Small hex screw in the wheel hub

During winter tires swap I've found this small screw loose. All other wheels have as well just 1 screw. Is there really ment to be just 1 screw or are all the others already missing? Whats the meaning of this screw?

It's to keep the rotor in place during assembly. There is only 1 per rotor. Tighten or remove it and you'll be good to go.
 
That looks great but at 20mm it's pushing it for me. On my BMW M3 I only did 12mm and I thought that was a bit a much for safety.
You get any vibration with these spacers.
They may look a little "wide" but both the front and rear (20mm) are exactly flush with the fender arches.

Hmmm, not sure what you mean by vibration, but I haven't noticed anything negative in the four months I've been driving with them.
