Small Screen Issue and Removing screen cover

So the other day this little tiny mark appeared on my screen

Any ideas what it could be?

I've got a spiegan cover on it so I'm not sure it could be dust or something?

Which leads to my next question

When removing a screen cover what's the best way to do it without damaging the screen? Just so I can get in there and have a look

It's probably behind the actual display, I have a couple in my work monitor, they can get anywhere
